Cell expressing a receptor of interest are incubated with a novel monoclonal antibody targeting the protein. Binding is performed at 4 °C to prevent internalization. Then cells are transferred to 37°C to allow internalization. At designated time-points, cells are washed with ice cold PBS to stop internalization. The novel antibody is detected with a secondary antibody. Cells can also be fixed and permeabilized to detect the receptor antibody combination in the intracellular compartment. Samples left at 4°C for the same duration show much less intracellular staining. This novel monoclonal antibody triggers internalization of its receptor upon binding.
