Case Study: Using bio-layer interferometry (BLI)

In our article on WD40 proteins (Moody, Wolfe, et al., 2021), we used bio-layer interferometry to examine the interactions of the WD40 protein WDR5 with Win – motif SET1 (SET1win) peptides to determine their kinetic fingerprint and affinities. An Octet RED384 and Strepavidin (SA) sensors were used for BLI, and a 3-fold dilution of WDR5 was conducted in ranges from 0.1uM to 9uM. The binding curves generated were made in ForteBio. Our results for this study demonstrated slow association and dissociation constants between our targets, providing a basis for the development of therapeutic agents targeting the Win binding site.
